Tutor:
Kateřina Dunovská, Martina Turchichová
Supervisor:
Department of Biomedical Technology
Synopsis:

Clinical laboratory instrumentation introduces principles of bioanalytical methods used in clinical diagnostics. Emphasis is put on optical methods (UV-VIS spectrophotometry, IR spectroscopy, AAS, AES, fluorimetry), NMR and X-ray analysis, electrochemical and electromigration methods (ion electrodes, biosensors, electrophoresis, isoelectric focusing), imunoassays and genetic methods (ELISA, PCR) as well as on chromatography and mass spectrometry. Contribution of lab automation to clinical diagnostics will be also discussed.

During the laboratory course students will be introduced into the basics of work in bioanalytical laboratory and lab data processing.

Requirements:

Assessment requirements:

Active attendance at all tutorials. Students has to come in time and be well prepared. Absences are acceptable for serious reasons only, have to be apologized and recompensed. Passing entrance test at least for 50% for each tutorial; if not, the tutorial including the test will be recompensed on another date. One exercise can be replaced at most.

Working out assigned exercises and submiting laboratory reports in satisfying quality.

Examination conditions: Student has to pass a written test comprising the topics discussed during the lectures and tutorials, including assigned individual study. The exam is graded according to the ECTS scale, to pass the exam, the test must be evaluated at least at 50 %.

Syllabus of lectures:

1. Princles of bioanalytical methods and clinical diagnostic

2. Optical methods I

3. Optical methods II

4. Electrochemical methods

5. Electromigration methods

6. Genetic techniques and imunochemical methods

7. Biosensors, automation in a clinical lab, POCT

8. Membrane processes, centrifugation and extraction

9. Chromatography

10. Mass spectrometry

Syllabus of tutorials:

1. Laboratory safety rules, basics of pipetting

2. UV-VIS spectrophotometry - quantification of proteins

3. Potentiometric titration of aminoacids and determining of water purity using conductivity measurement

4. Gel chromatography and simulation of HIV detection by ELISA

5. Excursion to a clinical laboratory of FN Motol

Study Objective:

Goal of the study is to get to know the principles of common bioanalytical methods used in a clinical laboratory, to work with scientific text and to discuss the current topics, and mainly, to solve model laboratory problems and tasks, which may happen in clinical diagnostics.
