Synopsis:

Structure and function of nucleic acids DNA and RNA. Replication, transcription, translation. Protein synthesis, prokaryotic and eukaryotic gene expression. Structure and function of proteins. Enzymes. Reproduction of cells, cell cycle, cell division. Biotechnology, hybridoma technology. Recombinant DNA vectors, restriction enzymes. Changes in genetic information, mutation. Methods of molecular biology - DNA isolation, centrifugation, electrophoresis, PCR. Flow cytometry. Genetic manipulation - genetic engineering, gene modification, gene splicing.

Requirements:

Credit: 80% attendance, laboratory exercises development, successful passing of 2written knowledge test

Exam: oral examination.

Caution: intervals between repeated examinations must not be shorter than seven days.

Syllabus of lectures:

1.Structure and function of DNA (the structure of the purine and pyrimidine bases, nucleotides, primary, secondary and tertiary structure of DNA, replication and DNA repair, DNA - the carrier of genetic information, gene structure, chromosome morphology, karyotype).

2.The central dogma of molecular biology, replication, bacterial genome, eukaryotic genome.

3. Structure and function of RNA (purine and pyrimidine bases, ribosomal RNA, messenger RNA, transfer RNA, the ribosome structure and function)The process of transcription, the complexity of eukaryotic genes post-transcriptional adjustments

4.4th The process of translation, gene expression and its regulation.

5.Structure and function of proteins, the physico-chemical properties, functions in the organism, cell exporting proteins, immunoglobulins - structure and function

6.Enzymes, enzyme-catalyzed reactions in the cell in the body.

7.Analysis of DNA. Isolation of DNA. Centrifugation. Electrophoresis.

8.Recombinant DNA restriction enzymes, PCR - reaction.

9.Changes in genetic information - mutation, mutagens, tumor transformation.

10.Flow cytometry.

11.Cloning of DNA vectors.

12.Biotechnology training of selected proteins. Hybridoma technology. 13.Genetic engineering. Transgenic organisms - GMOs.

14.Using molecular biology and genetics in clinical medicine. Gene therapy.

Syllabus of tutorials:

1. Safety at work in molecular-biological laboratory.

2.-4. Principles of sterilization techniques, working in a sterile box, preparation of cultivation medias.

5. Proteins and enzymes in the cell

6. -7. Biochemical analysis of DNA, isolation, centrifugation of samples.

8. -10. Identification of genes of Rh factor by PCR method., two master technician electrophoresis, application samples, readout and evaluation.

11.-13. Electropforesis of proteins. Methods of ELISA PCR

14. credit test

Study Objective:

To introduce fundamentals of molecular biology and to explain basic methods and applications of molecular biology.
