Synopsis:

Introduction to the structure and function of the optical microscope; familiarity with the technique of observation and drawing objektů.Nativní biological preparations, types of dyeing, the most commonly used dyes. Roztlakové medication. Permanent preparations, types of potting media, making permanent preparations. Preparation of samples for electron microscopy.

Requirements:

1st Mandatory participation in seminars. Non-participation is possible only for serious reasons such as illness, must be excused and exercise must be replaced.

2nd Of all laboratory exercises, the student must independently develop good quality protocol (see requirements for protocol) in laboratory workbook.

3rd When checking drawings of exercises oral interview, or writing to demonstrate understanding of the principle of each type of preparation and a description of the observed objects.

1st - Microscope. Construction, adjustment, operation.

2nd - Microscopic preparations. Observations made of native specimens.

Vital staining. Observation of mortality of cells.

3rd - Plasmolysis of cells and chemotaxis.

4th - Microscopic of prokaryotic and eukaryotic cells in hay infusion.

5th - Inverted microscope. Observation of cells in the phase contrast.

6th - Observations permanent preparations of animal tissues

7th - Cell division - mitosis

8th - Preparation of permanent slides

9th - Preparation of specimens and observation images of TEM, SEM

or excursion

10th - Preparation of specimens and observation omages of fluorescent

microscopy.
